Vimentin is one of the 5 major intermediate filaments found in cells (cytokeratin, vimentin, neurofilament, desmin, and glial fibrillary acid protein [GFAP]). It was classically thought to be limited to mesenchymal cells only, and in the early days of immunohistochemistry, it was thought to be a specific marker for sarcomas and other mesenchymal neoplasms.

We investigated CD99 expression in the development of normal ovary and testis as well as in 25 sex cord-stromal tumors (SC-STs), 7 epithelial neoplasms ab8069 staining Vimentin in human epithelial ovarian serous adenocarcinoma cell line SKOV-3 by Immunocytochemistry/ Immunofluorescence. Samples were fixed with 4% PFA in PBS pH 7.4 and then permeabilised using 0.2% saponin for 30 minutes. A blocking step was performed using 1% BSA/PBS for 1 hour. This antibody is intended for use to qualitatively identify vimentin by light microscopy in sections of formalin-fixed, paraffin-embedded tissue on a Ventana automated slide stainer.

Summary and explanation Vimentin is a 57 kDa intermediate filament (IF) protein, which form part of the cytoskeleton of vertebrate cells. Among the IFs, vimentin belongs to class III, showing a high degree of specificity for cells of mesenchymal origin. See below. Intracellular Staining by Flow Cytometry.

It can be used as a target for immunohistochemistry to help characterise numerous tissues and tumours. Invitrogen Anti-Vimentin Monoclonal (V9), eBioscience™, Catalog # 11-9897-82. They are paucicellular, have a myxoid stroma and thick-walled blood vessels. They typically stain for ER, PR, vimentin and desmin. Immunohistochemically, 239 poorly differentiated adenocarcinomas with solid components of the stomach were stained for vimentin. Vimentin‐positive cases were also stained by CAM 5.2 using serial mirror sections. We found 15 (6.3%) vimentin‐positive cases. Twelve of them demonstrated varying amounts of rhabdoid‐like cells.

The lymphoepithelium covering the bronchus-associated lymphoid tissue (BALT) of the rabbit lung was studied with monoclonal antibodies against vimentin, using the indirect immunoperoxidase technique.

Three alcohol-fixed lung cancers and two blocks of alcohol-fixed pleura failed to stain for vimentin. By contrast, all mesotheliomas and carcinomas, whether alcohol or formalin fixed, as well as sections of pleura, were strongly positive when stained with anticytokeratin antibody 35 beta H11.
